Description

Megaclox® is produced by Cipla.

Megaclox® comprise Ampicillin and Dicloxacillin as raw materials. Ampicillin is an extended-spectrum 3nd generation Aminopenicillin (drug class Penicillins). Ampicillin is beta-lactamase sensitive. Dicloxacillin is a β-lactamase (penicillinase)-resistant antibiotic of Penicillin class.

Indications:

- upper and lower respiratory tract infections (otitis, tonsillitis, bronchitis, throat and nose infections);

- gastrointestinal infections (enteritis, typhoid and paratyphoid);

- urogenital tract infections (urethritis, cystitis, pyelonephritis, prostatitis);

- skin and soft tissue infections (furunculosis, abscesses, cellulitis and skin ulcerations);

- bone, joint infections;

- meningitis;

- prophylaxis of bacterial endocarditis or abscess after minor surgery such as tonsillectomy, tooth extraction, as well as those who have changed their heart valves.

MOA (mechanism of action):

Ampicillin is a cell-wall inhibitor, that kills the bacteria by attacking the cell wall. Namely, it hinders the synthesis of peptidoglycan, which is vital for survival of bacteria in the human body. Dicloxacillin in its turn binds to beta-lactamases and inactivates them, thereby protecting Ampicillin, that is normally substrate for these enzymes. Thus, the activity of Ampicillin is enhanced if used in conjunction with Dicloxacillin.

Ampicillin acts as antimicrobial agent against Listeria monocytogenes, Escherichia coli, Salmonella typhi, Haemophilus influenzae, Streptococcus pneumonia, Streptococcus pyrogenes, Neisseria meningitidis, Proteus mirabilis, and Enterococcus faecalis.

Dosage

Megaclox® capsules should be taken orally on an empty stomach, at least 30 minutes to one hour before meals.

Duration of the therapy for common bacterial infection lasts for up to 14 days. This should be discussed with your doctor.

Adults and elderly

For infections of ear, nose and throat, bronchitis: 250 mg four times daily.

This may be increased to 1gram four times daily in certain cases.

For pneumonia: 500 mg four times daily.

For urinary tract infections: 500 mg three times daily.

For gastrointestinal infections: 500-750 mg three to four times daily.

For typhoid fevers:

Acute: 1-2 g four times daily for 2 weeks.

Carriers: 1-2 g four times daily for 4 to 12 weeks.

For Gonorrhoea: 2 g orally with 1 g of probenecid as a single dose. A repeated dose may be required in women.

The dosage may be reduced in patients with severe kidney failure.

Children under 10 years (20 kg or less)

For genitourinary or gastrointestinal tract infections: 100 mg/kg/day total, 4 times daily in equally divided and spaced doses.

For respiratory infections: 50 mg/kg/day total, in equally divided and spaced doses 3 to 4 times daily.

Missing a dose

If you miss a dose you should take it as soon as you remember about your missing. If it is the time for the next dose you should continue your regular dosing schedule. Do not take a double dose of Megaclox® to compensate the missed dose.

Overdose

If an overdose occurs and you are not feeling well, stop taking the medication immediately. You should seek emergency medical attention or contact your healthcare provider immediately.

Storage

Store at room temperature between 15 and 30 degrees C (59 and 86 degrees F) and away from excess moisture and heat. Throw away any unused medicine after the expiration date. Keep out of the reach of children.

Side effects

The most common side effects associated with Megaclox® are:

  • antibiotic-associated diarrhea
  • allergic reactions
  • nausea
  • vomiting
  • swelling of the lips, cheeks, tongue and eyelids
  • skin rash, itching
  • yellowing of the whites of the eyes and skin
  • discomfort in passing urine
  • blood in the urine
  • lower back pain

Side effects occurrence does not only depend on the medication you are taking, but also on your overall health and other factors.

Contraindications

Do not take Megaclox® if you are allergic to Ampicillin, Dicloxacillin or to other Penicillins.

Do not take Megaclox® if you are allergic to any other ingredient comprised in Megaclox®.

Do not take Megaclox® if you have a history of severe allergic reactions associated with using antibiotics.

Do not take Megaclox® if you have kidney problems.

Do not take Megaclox® if you have you have mononucleosis.

Do not take Megaclox® if you have if you have a stomach infection or diarrhea.

Do not take Megaclox® if you suffer from glandular fever or cytomegalovirus (CMV) infection.

Do not take Megaclox® if you suffer from lymphatic leukemia.

Do not take Megaclox® if you take other medications, including Allopurinol, sulfinpyrazone or probenecid (for gout); Tetracycline, Erythromycin (Erythro®) and chloramphenicol; anticoagulants (e.g. Warfarin) or phenindione; cytotoxic drugs e.g. Methotrexate (for rheumatoid arthritis, psoriasis, or lymphatic leukaemia); Chloroquine (for prevention and treatment of malaria); oral typhoid vaccine (for preventing typhoid).

Do not take Megaclox® if you are pregnant, plan to have a baby or are breastfeeding.
